Will a coil on plug work on a Guzzi?

The main issue would be to find a suitably sized coil, length, diameter to suit the reach required to seat on the plug.
May be applicable to some of the later model where the spark plug is in the centre of the combustion chamber and has an extended plug cap through the valve cover.
Probably not doable on the older tonti models where the plugs arent central. The standard coil, lead sparkplug in these is a proven and more reliable answer.

What model you thinking?  On older ones you'll have to make a mount on the bottom cover bolts(valve cover)  All the newer than 05 bikes cept Cali has a cover over the plug access (holes). How would you do twin plugs?

10mm plug on the inside, 14mm on the out side, remote coils, sparkplug leads, plug caps.
Absolutely no room between the inner plug and the tank for a coil on plug setup.

  Seems hard to imagine getting useful advice when you haven't mentioned what year and model of Guzzi you're talking about ?

2008 1200 Norge motor in a 1972 Eldo frame. I am mounting coils . My thought was locating the coils under the tank inside that long frame rail. It would require modification and be solid mounted. I was told that was a bad idea about the solid mounting. My next thought is coil on plug, lots of room.

you need to consider the coil ohms required by the  ignition. Don't want to solid mount coils ? Didn't the older Guzzis have coil brackets bolted to the frame? it's easy to rubber mount coils when they are not in plain sight..If you have 100 bucks to spend you may find modern micro sized. coils easier to mount..

Thats just CrAzY talk.  All my bikes are solid mounted, too many cars to count with solid mounting, Chevy used to solid mount right onto the intake manifold (now that is crazy)..
Best thing you can do for your coil is keep it away from heat. (and heat is a relative term).

My V700 has coil mounted solid on the bottom of said frame rail in the breeze. For 2 double output coils, put on a piece of flatstock bolted to framerail under it in the breeze.
